The township of Manchester was laid out in 1742 by Thomas Cookson. Mr. Cookson was a representative of the Lancaster County Courts, because at that time York County was a part of Lancaster County. The township originally extended from the Susquehanna River, to the Bermudian Creek, from the Conewago Creek, and to the Springetts Manor.

Manchester Township derived its name from the Duke of Manchester in England. It currently makes up 15.9 square miles of York and as of a 2000 census, served as a home for an estimated 12,700 residents.
